Aseptic Operation
Sterilization of the Sterile Room:
1. Regular cleaning of the sterile room should be done once a week. Start by mopping the floor and wiping the table and workbench with tap water. Then use 3% Sol, chlorhexidine, or 0.5% peracetic acid for final disinfection.
2. CO2 incubator (incubator) sterilization: Wipe with 3‰ Xinjieer, then with 75% alcohol or 0.5% peracetic acid, followed by UV irradiation.
3. Pre-experimental sterilization: Turn on the UV lamp, three-oxygen sterilizer, and air purifier system for 20–30 minutes each.
4. Post-experiment sterilization: Wipe the clean bench stage, side table, and inverted microscope with 75% alcohol (or 3‰ Xinjieer).
Aseptic Preparation of the Experimenter:
1. Wash hands thoroughly with soap.
2. Wear a lab coat, isolation cap, mask, and slippers.
3. Wipe hands with a 75% alcohol cotton ball before entering the sterile area.
Demonstration of Aseptic Operation:
1. All bottles containing alcohol, PBS, medium, and trypsin brought into the clean bench should be wiped with 75% alcohol on the outer surface.
2. Operate close to the alcohol lamp flame to maintain sterility.
3. Ensure all vessels are sterilized before use.
4. Utensils such as caps and pipettes should be kept in a high position and sterilized when in use.
5. Keep all operations near the alcohol lamp, perform actions gently and accurately, avoiding any unnecessary contact. Ensure that pipettes do not touch the waste container.
6. When handling multiple liquids, replace the pipette to avoid cross-contamination.
Cleaning and Disinfection of Instruments
Glassware Washing:
1. New glassware should first be washed with tap water to remove dust, then dried and soaked in 5% dilute hydrochloric acid for 12 hours to remove impurities.
2. After soaking, rinse with tap water, wash with detergent, and dry in an oven.
3. Soak in a cleaning solution (potassium dichromate 120g, concentrated sulfuric acid 200ml, distilled water 1000ml) for 12 hours, then rinse with tap water 15 times and distilled water 3–5 times.
4. Dry and wrap in kraft paper, then autoclave at 15 lbs for 20–30 minutes.
5. After autoclaving, dry the items in an oven.
Old Glassware:
1. Clean with Sur liquid or detergent solution, rinse, and dry.
2. Soak in cleaning solution for 12 hours, then rinse with tap water immediately and distill water.
3. Dry, wrap in kraft paper, and autoclave at 15 lbs for 20–30 minutes.
Metal Instrument Cleaning:
Wash with detergent, rinse with tap water, wipe with 75% alcohol, then rinse with distilled water and dry. Store in an aluminum box and autoclave at 15 lbs for 30 minutes.
Rubber and Plastic Items:
1. Wash with detergent, then tap water and distilled water, and dry.
2. Needle filter caps can be soaked in NaOH for 6–12 hours or boiled for 20 minutes. Install two membranes, ensure the glossy side faces up, and sterilize in an autoclave at 15 lbs for 30 minutes.
3. Rubber stoppers should be boiled in 2% sodium hydroxide for 30 minutes, then rinsed and treated with hydrochloric acid.
4. Plastic tips can be soaked in 75% alcohol for 5 minutes and exposed to UV light.
5. Plastic culture bottles and plates can be disinfected using UV or ethylene oxide, but ethylene oxide requires 2–3 weeks to dissipate.
Other Disinfection Methods:
Some items cannot be autoclaved. They can be immersed in 70% alcohol or disinfected with UV light. Ethylene oxide is also an option, but it must be fully removed after disinfection.
Preparation of Indicator Ink:
Mix 2g of cobalt chloride (CoCl₂·6H₂O), 10ml of 30% hydrochloric acid, and 88ml of distilled water. The ink becomes visible when heated, indicating successful disinfection.
Precautions:
1. Always check the pressure cooker for distilled water before autoclaving to prevent overheating. Ensure the safety valve is open to avoid explosions.
2. When installing filters, make sure the smooth side faces upward for proper filtration.
3. Wear acid-resistant gloves during acid treatment. Avoid splashing acid onto the floor or skin.
Subculture Cell Culture Notes:
1. Maintain strict aseptic conditions throughout the process.
2. Digest cells carefully. Stop digestion when cells begin to detach or float.
Cell Recovery:
The principle of cell resuscitation is rapid melting. Frozen cells at -196°C must be thawed quickly at 37°C to prevent ice crystal damage.
EDTA Digestive Solution Formula:
EDTA 0.20g, NaCl 8.00g, KCl 0.20g, KHâ‚‚POâ‚„ 0.02g, glucose 2.00g, 0.5% phenol red 4ml, and distilled to 1000ml. Autoclave at 10 lbs for 20 minutes. Adjust pH to 7.4 before use. Note: EDTA cannot be neutralized by serum. Thoroughly clean the flask after use to prevent cell detachment.
